CTLs from HLA-A2+ healthy donors were tested for epitope-specific cytotoxicity. CTLs with cytotoxic activity against unique peptide/HLA-A2 complexes were detected in 10 out of 11 different HLA-A2 healthy donors post expansion. Lung cancer cell lines were then tested as targets. CTL lysis of lung cancer cells with serine protease co-incubation was observed in 9 out of 10 confirmed expansions. CTL was measured using flow cytometry.
