This overall project, Linking community and food-web approaches to restoration: An ecological assessment of created and natural marshes influenced by river diversions, funded by the NOAA RESTORE Science Program, investigates how river diversions influence the living communities, food web structure, and function of created/restored and natural marshes in Louisiana to inform the development of marsh restoration strategies. Samples were collected in 2018, 2019, and 2021 from water, soil, and subtidal sediment from four natural marshes near Lake Hermitage, West Point a la Hache, and Bay Batiste, Louisiana, and two created/restored marshes within the Plaquemines Parish Lake Hermitage Marsh Creation Project (LHMCP). The LHMCP land was created from 2012-2014. were. Samples were taken in transects up to 100 m inland from the shoreline. Sediment and soil were collected from push cores up to 10 cm and sectioned in the field into separate depth intervals. From extracted DNA, the V4 region of 16S rRNA genes for bacteria and archaea were obtained using lotus-specific PCR primers and sequencing on the Illumina MiSeq platform and eukaryotic compositions were determined from 18S rRNA gene sequences.
