Helicobacter pylori strain H390 was isolated from gastric mucosa of a patient with gastric ulcer and was resistance to amoxicillin (the minimal inhibitory concentration, MIC was 256mg/L). But after 18 months of preservation at -80 , MIC to amoxicillin of the recovered culture decreased to 0.5mg/L. Then we used the clone H390 after cryopreservation H390 as the initial culture and obtained a stable amoxicillin resistant isolate H390r (MIC 256mg/L) by contacting amoxicillin in vitro. The whole genome of the clone H390 after cryopreservation and H390r were sequenced to explore the possible mechanism of the unstable and stable amoxicillin resistance.
