miRNA microarray in the synovial tissue 14 days after anterior cruciate ligament transection (ACLT)

To investigate changes in miRNA expression in the synovial tissue, miRNA microarray was performed at day 14 after ACLT and revealed that several miRNA expressions were changed more than 1.5-fold. We chose to analyze the role of miR-21 in joint pain after ACLT because it had the highest signal intensity among the increased miRNAs. Quantitative PCR confirmed that miR-21 expression in synovial tissue was significantly increased 14 days after ACLT.
为探究滑膜组织中微小RNA(microRNA, miRNA)的表达变化,本研究于前交叉韧带切断术(Anterior Cruciate Ligament Transection, ACLT)后第14天开展miRNA微阵列检测,结果显示多种miRNA的表达量变化幅度超过1.5倍。鉴于在表达上调的miRNA中,miR-21的信号强度最高,我们选择分析其在ACLT术后关节疼痛中的作用。实时定量聚合酶链反应(quantitative PCR, qPCR)验证结果表明,ACLT术后14天,滑膜组织内miR-21的表达水平显著升高。
