To investigate the changes of gene expression in pancreatic ductal adenocarcinoma cells under persistent endoplasmic reticulum stress, we treated AsPC-1 cells with 30 nM thapsigargin (Tg) for more than 14 days (designated as AsPC-1-Tg). To investigate whether the effects of persistent ER stress on gene expression is permanent or reversible, we cultured AsPC-1-Tg cells in a medium without Tg (designated as AsPC-1-Tg-Re) for 4 days. In this study, we then performed gene expression profiling analysis using data obtained from high-throughput RNA-seq (include mRNA, lncRNA, circRNA and miRNA) of AsPC-1 (WT), AsPC-1-Tg (TG) and AsPC-1-Tg-Re (RE) cells. Three biological replicates of each sample were used for RNA-seq. Overall design: Comparative gene expression profiling analysis of RNA-seq data for AsPC-1 (WT), AsPC-1-Tg (TG) and AsPC-1-Tg-Re (RE) cells.
