Complete genome assembly of the first Streptomyces acidocola isolate from Malaysia using Nanopore long-read sequencing and Illumina polishing

Genomes of Streptomyces are typically large, GC-rich, and highly repetitive, making them difficult to resolve using short-read sequencing alone, which often yields fragmented assemblies and disrupts the structure of biosynthetic gene clusters (BGCs). For Streptomyces acidocola, a recently described species lacking a complete reference genome, this limitation has hindered accurate characterization of its genomic architecture and metabolic potential. In this study, we applied a hybrid sequencing strategy combining Oxford Nanopore long reads with Illumina polishing to generate the first complete genome assembly, where long reads enabled resolution of repeats and complex regions while short reads ensured high base accuracy.
