Chromosomal translocations cause various lymphoid cancers and are prevalent in CRISPR-Cas9-meidated gene editing. However, the fate of gene editing-induced translocations remains elusive in vivo. Here, we found that translocations in engineered T cells experience clonal expansion after transferring into recipients for three weeks and remains comparable level with edited T cell before transfer, arguing against the notion that translocations would be gradually lost during cell proliferation due to growth retardation.
