Here, we investigate the stoichiometry of incompletely modified sites in tRNAs and tsRNA from human cells lines (MZ-294 and SH-SY5Y) for their information content. Using a panel of RNA modification mapping methods, we assess the stoichiometry of sites that contain the modifications 5-methylcytidine (m5C) by Bisulfite sequencing, 2’-O-ribose methylation (Nm) by RiboMethSeq and 3-methylcytidine (m3C), 7-methylguanosine (m7G) and Dihydrouridine (D) by AlkAniline Seq. The different cell lines have also been exposed to angiogenin to assess its impact on modification profiles.
