Material basis in Evodia rutaecarpa before and afte wine-processed for treating dysmenorrhea based on spectrum-effect relationship

The study aims to investigate the material basis of Evodia rutaecarpa (ER) before and afte wine-processed in treating dysmenorrhea based on spectrum-effect relationship. UPLC-Q-TOF-MS was used to analyze the blood-absorbed components of ER and its wine-processed product. The dysmenorrhea rat model was established. The effect of treating dysmenorrhea was evaluated using the writhing times, writhing latency, tumor necrosis factor-α (TNF-α), prostaglandin E2 (PGE2) and prostaglandin F2α (PGF2α) as indicators. Grey relational degree and entropy weight method were applied to analyze the spectrum-effect relationship between blood-absorbed components and analgesic efficacy. The results showed that 12 common peaks were detected from the serum samples of different batches ER and its wine-processed product, and 5 of these common peaks were identified. ER effectively reduced the writhing times, prolongated the writhing latency (P< 0.01), downregulated the contents of serum PGF2α and TNF-α, and upregulated the level of PGE2 (P< 0.05 or P< 0.01). Wine-processed Evodia Rutaecarpa (PER) had better than ER (P< 0.01). The results of spectrum-effect relationship indicated that the correlation score of 7-hydroxycoumarin and limonin significantly increased after wine-processed, and show positive correlation with treating-dysmenorrhea effect after wine-processed. This study clarified initially that the pharmacodynamic material of ER before and after wine-processed in treating dysmenorrhea, which provided an experimental basis for the clinical application of PER
