Chromosome Y sequencing of Han population in Henan, China

We used NGS to sequence 55 DNA samples from male Hans in Henan, China, to compare whether their haplogroups defined by Yleaf (Ralf, Montiel Gonzalez, Zhong, & Kayser, 2018) were in accordance with haplogroups defined by low-resolution methods(enzyme digestion, SSP, or SNaPshot). The samples were subjected to NGS using an Illumina HiSeq2000 platform (San Diego, CA, USA). A series of bait libraries was designed to capture the sequences of the ~11 Mbp region on the Y chromosome. We used previously described procedures for other steps prior to NGS, including DNA shearing, adaptor addition, gel electrophoresis, library preparation, bait design, and liquid-phase capture technology (Yan et al., 2014).
