Normal mouse development is regulated through dynamic changes in gene expression. Changes in RNA modification-related enzymes were confirmed using microarrays. Mouse embryos were selected at some development stages for RNA extraction and hybridization on microarrays. After inducing superovulation, natural mating was performed, and the day the vaginal plug was confirmed in the female mouse was designated as E0.5. Early embryos, including pronuclear (E0.5), 2-cell (E1.5), and morula (E2.5) stages, were collected and pooled. E10.5 and E16.5 embryos were collected whole-body from three individuals each.
小鼠正常发育进程受基因表达的动态变化调控。本研究借助微阵列(microarrays)技术,验证了RNA修饰相关酶的表达变化。研究选取多个发育阶段的小鼠胚胎开展RNA提取及微阵列杂交实验:通过诱导超数排卵后进行自然交配,将雌性小鼠检出阴道栓的当日记为胚胎发育第0.5天(E0.5);收集原核期(E0.5)、2-细胞期(E1.5)与桑椹胚期(E2.5)等早期胚胎并混合;分别从3只妊娠个体中完整采集E10.5及E16.5期的胚胎。
