Acute lung injury (ALI) is a severe inflammatory condition that disrupts pulmonary homeostasis, yet effective pharmacological treatments remain scarce. In this study, we applied a multi-dimensional strategy integrating chemical characterization, computational prediction, and in vivo validation to evaluate the therapeutic potential of a multi-component formula in juvenile rats challenged with lipopolysaccharide (LPS). Administration of the preparation ameliorated histopathological changes and inflammation in lung tissues, as evidenced by reduced levels of pro-inflammatory cytokines, and suppressed activation of AKT/TLR4/NF-κB signaling as well as the NLRP3 inflammasome. Parallel microbiome profiling indicated restoration of microbial community diversity, enrichment of beneficial bacterial taxa, and depletion of harmful genera. Correlation analyses further revealed that microbial shifts were associated with attenuation of inflammatory signaling. Collectively, these findings suggest that regulation of the gut–lung axis may contribute to the anti-inflammatory effects of SCP in the LPS-challenged model, primarily through modulation of inflammatory signaling.
