Taspase1 (threonine aspartase) is a 50kD endopeptidase of the family of proteases possessing an Asparaginase_2 homology domain. Loss of Taspase1 function in Taspase1 null (Tasp1-/-) mice led to craniofacial malformations. We used microarrays to determine which genes are affected by loss of Taspase1 during craniofacial development. E10.5 mouse embryo head (prosencephalon and mesencephalon) were dissected for RNA extraction and hybridization on Affymetrix microarrays. To identify which genes are regulated by Taspase1 during development, we used RNA from three wild type and three Taspase1 KO embryos.
Taspase1(苏氨酸天冬氨酸蛋白酶,threonine aspartase)是一种分子量为50kD的内肽酶,隶属于携带天冬酰胺酶2(Asparaginase_2)同源结构域的蛋白酶家族。在Taspase1基因缺失(Taspase1 null, Tasp1-/-)小鼠中,Taspase1功能丧失会引发颅面畸形。为明确颅面发育过程中受Taspase1功能缺失影响的基因,我们采用微阵列(microarrays)技术开展分析。我们对胚胎发育第10.5天(E10.5)的小鼠头部组织(前脑与中脑)进行解剖,以提取RNA并在Affymetrix微阵列芯片上完成杂交反应。为鉴定发育过程中受Taspase1调控的基因,我们使用了3份野生型胚胎与3份Taspase1基因敲除(KO)胚胎的RNA样本。
