In this experiment we performed STARR-seq on MDA-MB-231 cells that are treated with different drugs to test the features of cis-regulatory elements that make their activity potentially repressed by these small molecules. The STARR-seq library comprises fragmented genomic DNA of MDA-MB-231. The experiment contains four treatment groups: DMSO (control), CRY stabilizer SHP1705, proteasome inhibitor MG132, and the combination of SHP1705 and MG132. Prior to library transfection, media containing different drugs were added to cultured cell, and transfection mix was distributed to all groups. Eight hours after transfection, RNA was extracted, processed and sequenced.
