We investigated if the daily application of the interleukin-1 receptor-associated kinase (IRAK)-4 inhibitor PF-06650833 could rescue the effect of interleukin-1 beta on equine tendon cells cultured in 3D Overall design: Five lines of adult equine tenocytes (from different donors) were cultured in 3D in the presence of PF-06650833 alone, or IL-1ß plus PF-06650833 for 14 days. Gene expression profiles were generated by RNA sequencing on an Illumina Novaseq 6000. Quality control was perfromed using FASTQC and reads were aligned to the Equus Caballus transcriptome using the pseudoaligner Salmon. Differential expression was quantified using DESeq2.
