This study aims to investigate the role of the SARS-CoV-2 nsp15 protein in evading the host innate immune response. We generated recombinant SARS-CoV-2 variants (H234A and K289A) lacking nsp15 endonuclease activity and compared the innate immune response in human iPSC-derived alveolar type II epithelial cells infected with the wild-type virus versus nsp15 mutants through global transcriptomic analysis.
