Sequencing of Chlamydominas reinhardtii recombinant populations

In order to infer the distribution of fitness effects of new mutations we crossed mutation accumulation lines of the green alga Chlamydomonas reinhardtii with a mutation-free strain of the same genotype. The recombinant populations were then allowed to evolve under natural selection in the laboratory by serial transfer in liquid culture. We sequenced the recombinant populations at the beginning and after approximately 60 generations to estimate allele frequency changes and infer the distribution of fitness effects.
