DEtail-seq profiled meiotic DSBs in Saccharomyces cerevisiae

SPO11-promoted DNA double-strand breaks (DSBs) formation is a crucial step for meiotic recombination, and it is indispensable to detect the broken DNA ends accurately for dissecting the molecular mechanisms behind. Here, we report a novel technique, named DEtail-seq (DNA End tailing followed by sequencing), that can directly and quantitatively capture the meiotic DSB 3’ overhang hotspots at single-nucleotide resolution. The meiotic DSBs were strongly induced at 4 hours in synchronous meiosis, and enhanced when DMC1 was knocked out, but blocked when SPO11 was depleted. We performed DEtail-seq on the wild-type SK1 before meiosis (WT 0h), at 4 hours after meiosis (WT 4h), and DMC1 or SPO11 depleted samples at 4 hours after meiosis (DMC1KO 4h and SPO11 KO 4h, respectively).
