Deep amplicon sequencing of erg11/CYP51 of Leptosphaeria maculans

erg11/CYP51 is the target of the demethylase inhibitor (DMI) class of fungicides, and mutations at this locus can confer DMI resistance. In the canola pathogen Leptosphaeria maculans, promoter insertions at erg11 are known to increase tolerance to DMIs in Australia, but their prevalence across natural populations is unclear. To find novel alleles at erg11, as well as quantify genetic variation at this locus, we performed high-throughput sequencing of PCR amplicons of either the coding region ("C" samples) or promoter region ("P" samples) of erg11 derived from defined gDNA mixtures or natural ascospore populations.
