资源简介:
Atrial fibrillation (AF) following myocardial infarction (MI) represents a significant clinical challenge due to the lack of effective therapeutic options. However, the precise mechanisms underlying AF development remain poorly understood. This study systematically characterized the dynamic changes in differentially expressed genes associated with AF post-MI, thereby providing a foundation for a comprehensive molecular understanding of the disease. We investigated schemia-reperfusion –mediated gene expression alteration associated with the development of cardiac injury by microarray in a mouse model. All mice were euthanized via an overdose of pentobarbital on days 1, 3, and 7 post-myocardial infarction (MI). Total RNA was extracted from atrial tissues (n = 4 per group) at each time point using TRIzol reagent (Invitrogen) according to the manufacturer’s instructions. Gene expression profiling was conducted using the Affymetrix GeneChip Mouse Genome 430 2.0 Array following the manufacturer’s protocol (Affymetrix, Inc., Santa Clara, CA). The GeneChip Mouse Genome 430 2.0 Array contains over 45,000 probe sets that analyze the expression levels of more than 39,000 transcripts and variants derived from over 34,000 well-characterized mouse genes.
