To investigate the role of two secreted bacterial proteins (LphD and RomA) of Legionella pneumophila we infected THP-1 monocytes with GFP-producing bacteria. After 7 hours of infection, the cells were sorted (to enrich infected cells based on GFP-signal) and RNAseq was performed on the infected and sorted cells. Overall design: THP-1 monocytes were infected with either L. pneumophila wild type, ∆lphD, ∆romA, or ∆lphD∆romA. Non-infected cells were used as control and the resuls of the different knockout mutants were compared to wild type infected cells. All samples were analyzed in triplicate.
