During T cell activation hundreds of genes are primed for efficient activation. Here we show that IL-2 signaling is required to maintain open chromatin at gene regulatory elements which are associated with alternate pathways of T cell differetiation.IL-2 actiavtes binding of AP-1 and STAT5 at sites that can subsequently bind other STAT and lineage dietermining proteins, depending on what other factors exist in their enviroment. Overall design: DNase-seq samples were generated in duplicate for T-blast cells cultured in IL-2 or IL-7 with and without activation with PMA and calcium ionophore and in T-blast cells cultured for 16 hours in the absence of IL-2 or IL-7 (IL-2nil or IL-7nil) with and without activation with PMA and calcium ionophore. DNAse-seq samples were also generated from T-blast cells cultured with control DMSO or Ruxolitinib.
