The objective of the present study is to investigate the effect of endoplasmic reticulum (ER) stress caused by Hepatitis C Virus (HCV) infection on the mRNA expression profile in human hepatocytes. Overall design: Comparative gene expression analysis was performed using the cultured human hepatocellular carcinoma cell line Huh7.5.1. The cells were treated with 1 μg/mL tunicamycin for 48 hours to induce ER stress or infected with HCVcc (cell culture-derived HCV) for 48 or 72 hours. Gene expression was analyzed using 3D-Gene® Human 25k oligonucleotide microarrays, along with computational gene expression analysis tools to interpret the results.
