该数据面向研究细胞内蛋白质机器的结构和动态变化,发展了细胞水平的荧光寿命成像与分析技术,实现在细胞水平蛋白质机器的荧光标记,获得了细胞水平蛋白质的动态结构。该数据集包括包括细胞水平膜蛋白GPR17的荧光寿命成像数据,基于荧光寿命成像对GPR17同源二聚体建模获得的结构,野生型蛋白和突变体蛋白在膜环境分子动力学模拟过程中结构的变化曲线,GPR17野生型和突变体pulldown实验的原始图片及灰度分析。荧光寿命成像数据,pulldown实验在中国科学院精密测量科学与技术创新研究院实验平台采集,主要记录了GPR17蛋白在细胞内的荧光寿命变化。分子动力学模拟和结构建模数据在中国科学院精密测量科学与技术创新研究院计算平台的超算服务器上采集,主要记录了蛋白质结构中原子的空间坐标信息,数据量约为50MB
This dataset is developed for research on the structure and dynamic changes of intracellular protein machines. We developed cell-level fluorescence lifetime imaging and analysis techniques, achieved fluorescent labeling of protein machines at the cellular level, and obtained the dynamic structures of intracellular proteins.
This dataset includes: fluorescence lifetime imaging data of the membrane protein GPR17 at the cellular level; structures of GPR17 homodimers modeled based on fluorescence lifetime imaging data; structural change curves of wild-type and mutant proteins during molecular dynamics simulations in membrane environments; and original images and grayscale analysis results of pull-down assays for wild-type and mutant GPR17 proteins.
The fluorescence lifetime imaging data and pull-down assay data were collected on the experimental platform of the Innovation Academy for Precision Measurement Science and Technology, Chinese Academy of Sciences, which mainly record the intracellular fluorescence lifetime changes of GPR17 protein.
The molecular dynamics simulation and structure modeling data were collected on the supercomputing servers of the computing platform of the Innovation Academy for Precision Measurement Science and Technology, Chinese Academy of Sciences, which primarily document the spatial coordinate information of atoms in protein structures, with a total data volume of approximately 50 MB.
