Spongospora subterranea is a soil borne biotrophic pathogen that causes powdery scab of potato and vectors Potato mop-top virus (PMTV). Obtaining high-quality nucleic acids is difficult due to the durability of the sporosori, intracellular growth, and obligate life style of the pathogen. Genomic DNA was isolated from root galls collected from a field-grown Russet Burbank potato plant in Minnesota State, USA was sequenced by a Minion Nanopore Systems. Sequenced data were De Novo assembled and polished with short reads from previously sequenced isolate by Illumina Miseq. The newly polished assembled genome mapped 100% to the only published draft genome, SSUBK13. The genome was assembled into 346 scaffolds with an estimated size of 31.51 Mb and 10,325 protein-coding genes. The GC content is 45.65% and consistent with other sequenced Plasmodiophorids except Plasmodiophora brassicae. This genome resource will aid in improving our understanding of the genetic architecture of S. subterranea and development of new strategies for managing powdery scab of potato.
