In this study, we used the EMPIRIC approach of deep mutational scanning to estimate the selection coefficients of all 118 single amino acid mutations in the middle domain of Hsp90 in yeast populations across 5 different environments at normal expression levels. The Hsp90 middle domain plays a prominent role in client binding interactions, and was thus hypothesized to play an important role in the potential adaptive response under different environmental conditions. Altogether, our study sheds light on the potential role of specific protein regions during adaptation to new environments and the variation of the DFE along the genome.
