资源简介:

Background. High-oleic palm oil (HOPO) from interspecific O×G hybrids (Elaeis oleifera × Elaeis guineensis) is enriched in oleic acid and bioactive minor components (tocotrienols, tocopherols, carotenoids, phytosterols and phenolics) with antioxidant activity. However, in vivo evidence of its biological effects in models of neurodegeneration-associated proteotoxicity is limited. Objective. To evaluate the effects of a HOPO bioextract on reproductive performance, survival and locomotor activity in wild-type and transgenic Caenorhabditis elegans models of Alzheimer’s and Parkinson’s disease proteotoxicity. Methods. Wild-type N2 and the transgenic strains CL2006 (muscle Aβ1–42) and NL5901 (muscle α-synuclein::YFP) were exposed to a HOPO bioextract at 6.25, 12.5 and 25 µg/mL and compared with vehicle-matched controls. Reproduction and locomotor activity were modelled with generalized linear mixed models (negative-binomial and Poisson distributions, respectively), with estimated marginal means and Tukey-adjusted pairwise contrasts. Survival was analysed by Kaplan–Meier estimation with log-rank tests, and hazard ratios were obtained from Cox proportional-hazards models. Results. The bioextract did not significantly modify any endpoint in N2. In both transgenic strains, reproduction and survival improved significantly relative to controls in a concentration-dependent manner; at 25 µg/mL, the bioextract reduced the risk of death by approximately 76–77% (CL2006 HR = 0.24; NL5901 HR = 0.23). Locomotor activity improved at 25 µg/mL in CL2006 and in a concentration-dependent manner in NL5901. Conclusions. The HOPO bioextract improved reproduction, survival and locomotion in C. elegans models of proteotoxic stress, most markedly in the NL5901 model, supporting further evaluation of its bioactives for healthy-aging and nutraceutical applications. Contents. Raw data for reproduction (cumulative progeny), survival (daily scoring over 15 days) and locomotion (body bends), characterization data of the bioextract, and the R scripts used for the statistical analyses reported in the associated manuscript.
