Due to overwintering of M. wufengensis, it is important to dissect the gene network for anthocyanin biosynthesis during different flower color changing stages. To this end, a de novo assembly RNA-seq method was employed to analyze the petals of M. wufengensis from five flower development stages. Surprisingly, a R2-R3 type MYB transcription factor (we named it MwMYB1) was found. Expression of MwMYB in tobacco or Arabidopsis results in red color leaves, which infers that MwMYB is a key player in anthocyanin biosynthesis. In addition, MwMYB works independently in controlling anthocyanin biosynthesis related gene expression. Taking advantage of DAP-seq analysis, MwMYB-1 prefers AAGAGAG cis-element in MYB75 gene to regulate its expression, which robustly helps accumulate anthocyanin content in leaves or flowers. Furthermore, this study will provide evidence for molecular breeding concerning flower color modifications in tree species.
