Diel transcriptome of Lemna aequinoctialis under three photoperiod conditions

To investigate the molecular mechanisms under photoperiodism, the diel transcriptome of a short-day flowering duckweed, Lemna aequinoctialis (synonym: L. paucicostata, L. aoukiuksa) was obtained. As a material, the Nd strain (OYA0001 strain) whose critical day-length is ca. 13 h was used. Plants were precultured under 15L9D and sampled every 2 h in a day after the cultivation under three photoperiod conditions, 14L10D (one week), 12L12D (one week) and 9L15D (two weeks). These date will provide the phase information of circadian clock genes and their response to the day-length change. The floral development genes will be detected by comparing the gene expression between long-day and short-day conditions. The mRNA was extracted from a whole-plant sample by magnetic beads. The library was prepared by BrAD-seq protocol with shot-gun primers. The 150bp PE sequences were performed by Hiseq system.
