Gene expression array of brain, mandible and maxilla tissues from P0 FoxO6-/- and wildtype mice

FoxO6 is expressed in the brain, craniofacial region and somite, but the precise role of FoxO6 in craniofacial development remain unknown. We found that FoxO6 is expressed specifically in craniofacial tissues and FoxO6-/- mice undergo expansion of the face, frontal cortex, olfactory component and skull. Our gene expression array data demonstrated that Lats1, an important component of Hippo signaling pathway, was decresed in FoxO6-/- mice. Our in vitro and in vivo data further validated that FoxO6 activates Lats1 expression, thereby increasing Yap phosphorylation and activation of Hippo signaling. 叉头框O6(FoxO6)在脑、颅面区域及体节中均有表达,但FoxO6在颅面发育中的确切功能尚未明确。本研究发现,FoxO6特异性表达于颅面组织中,且FoxO6基因敲除(FoxO6-/-)小鼠会出现面部、额叶皮层、嗅觉结构及颅骨的体积增大。我们的基因表达芯片数据显示,Hippo信号通路的重要组分大肿瘤抑制激酶1(Lats1)在FoxO6-/-小鼠中表达下调。后续体外与体内实验进一步验证,FoxO6可激活Lats1的表达,从而增强Yes相关蛋白(Yap)磷酸化及Hippo信号通路的激活。本研究使用出生后0天(P0)的FoxO6-/-小鼠与野生型(wildtype)小鼠的脑、下颌骨及上颌骨组织,开展Affymetrix基因芯片检测。
