Glomerular Transcriptome from Experimental Nephrotic Syndrome Model and Treatment with PPARgamma Agonists [ADR]

Nephropathy was induced with puromycin amino-nucleoside (PAN) in Wistar rats and treated with daily treatments of Pioglitazone (Pio) and GQ-16. Nephropathy was induced in a separate group of rats with Adriamycin (ADR) in Wistar rats. Total glomerular RNA was isolated, and mRNA libraries were generated and subjected to sequencing using NovaSeq6000 SP. Injury with PAN and ADR resulted in differential expression of genes (DEGs). Many of the PAN-induced DEGs were restored with Pio and GQ16 treatments.
本研究采用嘌呤霉素氨基核苷(puromycin amino-nucleoside, PAN)诱导Wistar大鼠构建肾病模型,并通过每日给药吡格列酮(Pioglitazone, Pio)与GQ-16进行干预。另设一组Wistar大鼠,采用阿霉素(Adriamycin, ADR)诱导构建肾病模型。提取各组大鼠的肾小球总RNA,构建mRNA文库,并利用NovaSeq6000 SP测序平台完成测序。PAN与ADR造模均可引发大鼠肾脏基因的差异表达(differential expression of genes, DEGs)。经吡格列酮与GQ-16处理后,多数由嘌呤霉素氨基核苷诱导产生的差异表达基因可得到恢复。
