Genetic variation modulates aberrant DNA hypomethylation in naïve pluripotent stem cells.

We observed markedly different DNA methylation chances in response to MAP kinase inhibition between 129 and B6J mESCs. We reasoned that these differences at least in part are driven by the differential expression of DNA methylation regulators and conducted RNA sequencing experiments. Overall design: Mouse ES cells of B6J or 129 background were cultured in complete ES cell media with serum/LIF.DMSO or 1uM of Mek inhibitor (iMEK-PD 0325901) was added to the culture from Day 0 to Day 5 daily. RNA was extracted at the end of 5 days of treatment and processed for RNA sequencing.
