Nuclei were isolated from left ventricular heart tissue harvested from control (healthy) and hypertrophic individuals. Human heart specimens were provided by the Department of Forensic Medicine in Stockholm, Sweden. Ethical permissions were obtained from local ethical committees. The myocardial left ventricles were collected after receiving the informed consent of relatives. After obtaining the tissue, left ventricle myocardial tissue was dissected, without the septum, snap-frozen and stored at -80°C until further processing.Cardiac myocyte nuclei were isolated using PCM-1 immunolabelling and flow cytometry assisted sorting, according to Bergman et al., 2012. Strand-specific RNA-sequencing was performed on 4 control (ctrl) and 4 hypertrophic (LV CH) human hearts.
