We isolated and cultured virus from the acute-phase serum of a patient with severe fever with thrombocytopenia syndrome (SFTS) and obtained an SFTSV strain. The viral culture supernatant was then sent to Hefei Jiakangming Biotechnology Co., Ltd. for metagenomic next-generation sequencing (mNGS). The main steps were as follows: (1) Sample preprocessing and nucleic acid extraction: the culture supernatant was centrifuged to remove impurities, total RNA was extracted using a commercial kit, and residual host DNA was removed by DNase I treatment; (2) Library construction and quality control: rRNA/host nucleic acids were depleted from the extracted nucleic acids, cDNA was synthesized by reverse transcription, followed by end repair, A-tailing and adaptor ligation to construct the sequencing library, whose concentration and fragment size distribution were assessed using Qubit and a bioanalyzer; (3) High-throughput sequencing: paired-end sequencing was performed on an Illumina high-throughput sequencing platform to obtain raw sequencing data; (4) Data processing and assembly: raw reads were subjected to adaptor trimming, low-quality filtering and deduplication, clean reads were mapped to the human reference genome to remove host sequences, and the remaining virus-related reads were de novo assembled and further polished with a reference genome, ultimately yielding a (near-)complete genome sequence of this SFTSV strain, which was exported in FASTA format for inclusion in this dataset.
