Influence of two bacterial effectors on the transcriptional landscape of the host cell

To investigate the role of two secreted bacterial proteins (LphD and RomA) of Legionella pneumophila we infected THP-1 monocytes with GFP-producing bacteria. After 7 hours of infection, the cells were sorted (to enrich infected cells based on GFP-signal) and RNAseq was performed on the infected and sorted cells. Overall design: THP-1 monocytes were infected with either L. pneumophila wild type, ?lphD, ?romA, or ?lphD?romA. Non-infected cells were used as control and the resuls of the different knockout mutants were compared to wild type infected cells. All samples were analyzed in triplicate.
