Firstly, a DMD monkey model with exon 50 mutation was constructed using CRISPR/Cas9 to analyze whether CRISPR/Cas9 caused off-target. Then, the gene repair of the targeted exon 51 was carried out, and the target efficiency and off-target were analyzed, so as to prove the safety of the therapeutic gene.
