Seasonal dynamics of bacterioplankton community in a large, shallow, highly dynamic freshwater lake

Water samples (0.5 m depth below water surface) were collected using a plexiglass water sampler in April (spring), July (summer) and October (autumn) 2014, and January 2015 (winter). At each sampling site, triplicate samples apart around 200-500 m were collected. A total of 48 samples were obtained. The collected water samples were then transported to the laboratory on ice within 24h. A sub-sample of water (500 ml) for the 16S rRNA gene analysis was filtered sequentially through 3 µm and 0.2 µm pore size membrane filters (polycarbonate, Millipore, USA) and these filters were frozen at -80°C until DNA extraction. Another set of sub-sample (500 ml) was used for physicochemical analysis.
