Methylome in human cells expressing mycoplasmal DNA methyltransferases

We expressed Mycoplasma hyorhinis MTases MhyI and MhyII in cultured human cells. To determine the genome-wide effect of MhyI and MhyII-induced methylation, we performed global methylation profiling of the genomic DNA isolated from HT1080/MhyI, HT1080/MhyII and HT1080/mock cells. We employed the genome-wide Methylated DNA ImmunoPrecipitation – genome sequencing (MeDIP-seq). To obtain the 5mC-enriched DNA fraction, we used an anti-5mC antibody (clone 10G4) that is highly specific to 5mC in both the CG- and GATC-methylated sequences. We sequenced DNAs in the MeDIP fraction and recorded over 10 Gb of sequence reads per sample.
