SPC2B2 iAT2s were transduced with either control WT YAP or constitutively active nuclear YAP (YAP5SA) lentivirus in a single cell suspension and were then grown in iAT2 media (CK + DCI) in 3D matrigel for 7 days. iPSCs (clone SPC2-ST-B2) underwent directed differentiation to produce lung alveolar epithelial type 2 cells (iAT2s), maintained in CK+DCI medium in 3D cultures. The iAT2s were then lentivirally transduced in a single cell suspension with either control WTYAP or constitutively active nuclear YAP (YAP5SA) lentivirus ((each vector also carrying a TagBFP reporter cassette), and the resulting cells grown out in 3D CK+DCI cultures were captured for profiling by scRNA-seq.
