An opportunistic intracellular pathogen Mycobacterium avium subsp. hominissuis, a member of the nontuberculous mycobacteria (NTM) cluster, causes respiratory disease in immunosuppressed hosts. In particular, infected companion dogs are a potential role to transmit the agent to children or immunosuppressed people. The purpose of this study is to investigate a host-M. avium hominissuis interactome in canine PBMCs during the infection. Peripheral blood mononuclear cells were isolated from buffy coats of six healthy beagle dogs by density gradient centrifugation. PBMCs were stimulated in vitro with M. avium hominissuis strain 104. Total RNAs were extracted from the cells at 2h, 6h, 12h, and 24h after infection. The construction of library was performed using QuantSeq 3’mRNA-Seq Library Prep Kit. High-throughput sequencing was performed as single-end 75 sequencing using NextSeq 500.
