Single-cell RNA-seq from primary somatosensory cortical S1L2/3glutamatergic neurons of the mouse

We analyzed the gene expression profiles of S1L2/3 glutamatergic neurons in maternal mice and their offspring by single-cell RNA sequencing (12,671 ± 1,402 genes per neuron). Compared to sham control mice, we identified 513 and 571 differentially expressed genes in CCI offspring and their maternal mice respectively. And 102 overlapped genes were identified between CCI offspring and their maternal mice. Single-cell RNA-seq using SMART-Seq2 method was performed from individual S1 glutamatergic neuron after patch clamp recording. We performed RNA-seq on total 9 neurons from CCI offspring, CCI maternal and sham mice and three replicated neurons for each group.
