Acid mine drainage of the Carnoules mine (France) is characterized by acid waters containing high concentrations of arsenic (50-350 mg/l) and iron (around 2 g/l). Along the Reigous, a small creek draining the site, more than 99% of the dissolved arsenic was removed by co-precipitation with Fe(III). This natural attenuation of arsenic and iron involved iron and arsenic oxidizing bacteria. Among arsenic oxydizing bacteria, Thiomonas genus were recognized to be an important population in this site. The bacterial community was characterized in sediments along this creek and were combined with a FISH study and a Selected Reaction Monitoring (SRM)-based proteomic approach developed to detect and quantify the two subunits of the arsenite oxidase and RpoA of two different Thiomonas groups. Pyrosequencing was performed by a Roche 454 Life Sciences Genome Sequencer FLX Titanium sequencer.
