Clavibacter michiganensis is a Gram-positive bacterium with eight subspecies, five of which have been redefined as species according to genomic data. Based on phylogenetic analysis with dnaA gene sequences, Clavibacter strains isolated from barley have been grouped in a separate clade from other Clavibacter species and subspecies. In this study, the biochemical, physiological, and genetic characteristics of strain DM1 and DM3, which represented the barley isolates, were examined. Based on multi-locus sequence typing and other biochemical and physiological features, including colony colour, carbon source utilisation, and enzyme activities, strain DM1 and DM3 were categorically differentiated from the above mentioned eight species and subspecies of Clavibacter. Moreover, genomic analysis showed that the DNA G+C content of strain DM1 was 73.7%, and the Average Nucleotide Identity values between DM1/DM3 and other species/subspecies are ranged from 89.0 to 91.5%. The ANI value between DM1 and DM3 are 98.4%. These results indicated that DM1/DM3 was distinct from other known Clavibacter species and subspecies. Therefore, we proposed a novel species, C. hordeum, with the strains DM1 and DM3.
