PRLX-93936 is an optimized derivative of erastin. To determine the downstream effects of PRLX-93936 on gene expression, we performed mRNAseq of PANC-1 cells treated with the drug or vehicle control. Overall design: PANC-1 cells were seeded in 6-well plates. The following day, cells were treated in triplicate with 0.5 uM PRLX-93936, 5 uM PRLX-93936, or DMSO vehicle control for 2 hours and 6-hours. RNA was isolated using RNeasy Mini Kit (QIAGEN) with DNase treatment. RNA QC, library preparation, and Novaseq sequencing were performed by Genewiz. Gene-level expression was quantified from RNA-seq using the nf-core/rnaseq pipeline (version 3.12.0). Salmon (version 1.10.1) was used to generate gene-level read counts.
