Using as a model the German cockroach, Blattella germanica, we examined the activity of AGO2 by depleting its mRNA. A second treatment with a dsPolyh (and heterologous dsRNA) allows the analysis of the siRNAs generated from this dsRNA, in control and AGO2-depleted animals. The use of a heterologous dsRNA (dsPolyh) allowed us to dissociate the activity of dsRNA processing and selection of the guide siRNA from the action of cleavage a complementary RNA.
