An opportunistic intracellular pathogen Mycobacterium intracellulare, a member of the nontuberculous mycobacteria (NTM) cluster, causes respiratory disease in immunosuppressed hosts, including companion dogs. The purpose of this study is to investigate a host-M.intracellulare interactome in canine monocyte-derived macrophages during the infection. Overall design: Monocyte-derived macrophges (MDMs) were isolated from buffy coats of ten healthy beagle dogs by density gradient centrifugation. MDMs were stimulated with M. intracellulare. Total RNAs were extracted from the cells at 0, 6, 24, and 72h after infection. The construction of library was performed using QuantSeq 3'mRNA-Seq Library Prep Kit. High-throughput sequencing was performed as single-end 75 sequencing using NextSeq 500.
