We develop a sprayable biomimetic double mask (BDM) with rapid autophasing and hierarchical programming for scarless wound healing. The BDMs are photocrosslinkable and comprise two distinct layers with a hydrophobic top layer composed of poly (lactide-co-propylene glycol-co-lactide) dimethacrylate (PGLADMA) polymer, and a hydrophilic bottom layer consisting of gelatin methacrylate (GelMA) hydrogel. Upon mixing, these two phases undergo spontaneous water/oil separation, enabling swift autophasing into bi-layered structure. After photocrosslinking, the BDMs could be rapidly solidified with strong interfacial bonding, robust tissue adhesion, and excellent joint adaptiveness. Upon implementation, the BDMs hierarchically program the dynamic healing cascade both in vitro and in vivo. The bottom GelMA layer massively release Ca2+ for rapid hemostasis, while the top PGLADMA layer maintain a moist and sterile environment, synergistically suppressing the inflammatory TNF-a pathway to advance the transition from inflammation to proliferation. Our BDMs also coactivate and coordinate the cGMP/PKG-Wnt/Ca2+ signaling pathways to nourish the vascular reconstruction. Collectively, our BDMs possess huge transformative potential for scarless wound healing and offer substantial benefits to patients afflicted by extensive wounds.
