This project involves understanding the genetic determinants of trimethoprim resistance in Escherichia coli using laboratory evolution. E. coli K-12 MG1655 was subjected to 300 ng/ml or 50 ng/mL trimethoprim in a serial passage model for experimental evolution. Resistant clones were isolated on1 microgram/mL trimethoprim. Resistance was also evolved in a Lon-knockout background at 300 ng/mL.
