GMDS is important for human lung adenocarcinoma progression, detailed molecular mechanisms remain elusive. We used microarray to detail the global gene expression changes in A549 cells with GMDS knockdown as compared to control cells A549 cells infected with lentivirus expressing either Scr-shRNA (n=3) or GMDS-shRNA (n=3) were cultured and then total RNA was extracted using Trizol reagents. RNA quantity and quality were determined using NanoDrop 2000 and Agilent Bioanalyzer 2100. For gene expression analysis, Affymetrix human GeneChip primeview was used according to the manufacturer’s instructions. In brief, GeneChip 3’ IVT Expression Kit was used for first-strand complementary DNA synthesis, double-stranded DNA template conversion, in vitro transcription for aRNA synthesis and labelling. Microarray hybridization, washing and staining were done using GeneChip Hybridization Wash and Stain Kit.
