The protein kinase KIS impacts gene expression in the neonate brain

The protein kinase KIS (product of the gene UHMK1) is expressed during mouse brain development and is proposed to control gene expression through the phosphorylation of splicing factor SF1. We compared the expression of transcripts in wild type and KIS-ko newborn mouse brain (females) using affymetrix exon 1st arrays. Six pairs of wildtype and KIS-ko brains from littermate female newborn mice were processed for Affymetrix exon 1st array hybridization. Analysis was performed at the exon level using PLIER normalisation and the Affymetrix Expression Console.

蛋白激酶KIS(protein kinase KIS,基因UHMK1的编码产物)在小鼠脑发育过程中表达,被推测通过剪接因子SF1(splicing factor SF1)的磷酸化调控基因表达。我们采用Affymetrix外显子1st阵列,对比野生型(wild type)与KIS基因敲除(KIS-ko)新生雌鼠脑组织的转录本表达水平。共收集6对同窝新生雌鼠的野生型与KIS敲除脑组织样本,开展Affymetrix外显子1st阵列杂交实验。数据分析在外显子水平进行,采用PLIER标准化(PLIER normalisation)方法与Affymetrix Expression Console软件完成。
